چکیده
Xenocoumacin 1 (Xcn1), which is produced by Xenorhabdus nematophila CB6, exhibits strong inhibition activity against plant pathogens, especially fungi and oomycetes. Therefore, it has attracted interest in developing into a novel biofungicide applicable for protection. However, its low yield with concomitant high cost during the fermentation process limits widespread application. In this study, we replaced native promoter of xcnA arabinose-inducible araBAD (PBAD), well-known widely used expressing heterologous genes, to evaluate effects on Xcn1 antimicrobial activity. Compared wildtype strain, was improved from 68.5 mg/L 249.7 (3.6-fold) 234.9 (3.4-fold) at 0.5% 1.0% L-arabinose concentration, respectively. We further explored transcription level biosynthesis related genes found that their upregulation resulted improvement Xcn1. Moreover, Bacillus subtilis Phytophthora capsici determined agar diffusion plate growth assay, as expected, also be enhanced. The promoter-replacement strategy utilized here improves efficiently, provides basis industrial production

Xenorhabdus nematophila: Mutualist and Pathogen
T he gram-negative bacterium Xenorhabdus nematophila engages in a mutualistic relationship with a specific soil nematode and also can mount potent pathogenic attacks on a variety of insects. During its mutualisticpathogenic life cycle, Xenorhabdus produces a wide range of exoenzymes, antimicrobial and nematicidal compounds, and insect toxins. Moreover, the nematode-bacteria complex is useful fo...
متن کاملAnalysis of the PixA inclusion body protein of Xenorhabdus nematophila.
The symbiotic pathogenic bacterium Xenorhabdus nematophila produces two distinct intracellular inclusion bodies. The pixA gene, which encodes the 185-residue methionine-rich PixA inclusion body protein, was analyzed in the present study. The pixA gene was optimally expressed under stationary-phase conditions but its expression did not require RpoS. Analysis of a pixA mutant strain showed that P...

The Xenorhabdus nematophila nilABC genes confer the ability of Xenorhabdus spp. to colonize Steinernema carpocapsae nematodes.
Members of the Steinernema genus of nematodes are colonized mutualistically by members of the Xenorhabdus genus of bacteria. In nature, Steinernema carpocapsae nematodes are always found in association with Xenorhabdus nematophila bacteria. Thus, this interaction, like many microbe-host associations, appears to be species specific. X. nematophila requires the nilA, nilB, and nilC genes to colon...
متن کاملInsecticidal pilin subunit from the insect pathogen Xenorhabdus nematophila.
Xenorhabdus nematophila is an insect pathogen and produces protein toxins which kill the larval host. Previously, we characterized an orally toxic, large, outer membrane-associated protein complex from the culture medium of X. nematophila. Here, we describe the cloning, expression, and characterization of a 17-kDa pilin subunit of X. nematophila isolated from that protein complex. The gene was ...
